A practical reference on secretagogue: what it is, how it behaves, what the literature reports, and where the honest uncertainties sit.
Reviewed 2025-11-17. Anything still debated is marked as such rather than presented as settled.
The molecule contains five residues, including alpha-aminoisobutyric acid, D-2-naphthylalanine, and D-phenylalanine, and it ends in a lysine amide. Non-natural and D-configured residues make the chain less susceptible to common peptidases, which helps explain its resistance to rapid breakdown. Its molecular formula is C38H49N9O5, corresponding to a free-base mass near 711.9 daltons. The C-terminal amide removes a negative charge and is a recurring feature in receptor-active peptides of this family. These structural choices are usually discussed as the basis for its selectivity profile.
Published animal and early human work describes growth hormone release that is separated from comparable rises in adrenocorticotropic hormone and cortisol. Prolactin changes are reported as small in the same studies. Selectivity is attributed to binding at the ghrelin receptor and to the downstream signaling that follows, rather than to differences in how quickly the peptide is cleared. Authors commonly label the compound selective rather than potent, because the same mass produces a smaller growth hormone response than some older secretagogues tested in parallel. Whether that profile holds across species and routes of administration remains an open question.
Compared with older secretagogues such as hexarelin or GHRP-6, ipamorelin shows weaker stimulation of cortisol, prolactin, and appetite in the animal models used for early characterization. Whether that selectivity is preserved across longer human exposures remains an open question, because published clinical data are limited in size and duration. Reported effects on food intake are generally described as modest. The compound is therefore treated in the literature as a relatively selective research tool rather than a fully characterized therapeutic agent.
Ipamorelin is a synthetic pentapeptide that belongs to the growth hormone secretagogue class of compounds. Researchers at a pharmaceutical company first described it in the 1990s while screening small peptides for growth hormone releasing activity. Its chain contains five amino acid residues, two of which are non-natural building blocks, including 2-aminoisobutyric acid and a naphthylalanine derivative. The molecule was designed to act at the ghrelin receptor while avoiding several effects observed with earlier secretagogues.
At the cellular level, ipamorelin binds the growth hormone secretagogue receptor, also called the ghrelin receptor. Activation of this receptor on pituitary somatotroph cells triggers a signaling cascade that leads to release of growth hormone into circulation. Because release follows a pulsatile pattern, studies often report peak concentration and total area under the curve rather than a single time point. Selectivity for this receptor is the property most frequently discussed in comparative work.

At the molecular level, ipamorelin acts as an agonist at the growth hormone secretagogue receptor type 1a, the same G protein-coupled receptor that binds ghrelin. Receptor activation couples to Gq/11 signaling, raising intracellular calcium through inositol trisphosphate and diacylglycerol, which in turn promotes exocytosis of growth hormone from pituitary somatotroph cells. Ipamorelin binds this receptor with high affinity and shows weak activity at other secretagogue-related targets in vitro. Its action requires the intact receptor and is not reversed by growth hormone-releasing hormone antagonists.
Compared with earlier growth hormone secretagogues such as GHRP-6 and hexarelin, ipamorelin has been reported to produce less stimulation of adrenocorticotropic hormone, cortisol, and prolactin in animal and early human studies. This selectivity is usually attributed to differences in receptor subtype interactions and to the tissue distribution of the receptor. Effects on appetite appear weaker than those of ghrelin itself, although the supporting evidence base is small. Whether these differences produce a distinct clinical profile remains an open question, since controlled human trials are limited.
Ipamorelin is a synthetic pentapeptide first described in the 1990s by researchers at Novo Nordisk during a program to develop selective growth hormone secretagogues. Its sequence is Aib-His-D-2-Nal-D-Phe-Lys-NH2, incorporating two non-natural residues, alpha-aminoisobutyric acid and D-2-naphthylalanine. The C-terminus is amidated, and the material is supplied as a white lyophilized powder. The molecular formula is C38H49N9O5 and the monoisotopic mass is approximately 711.85 daltons. The short chain and modified residues give it greater resistance to enzymatic degradation than many larger peptide hormones.
Ipamorelin is a synthetic pentapeptide that belongs to the growth hormone secretagogue class of compounds. Its sequence is Aib-His-D-2-Nal-D-Phe-Lys-NH2, a structure that differs from natural ghrelin in length and in the presence of non-natural amino acid residues. Early laboratory work described it as a comparatively selective agent that stimulates growth hormone release with limited effect on other pituitary hormones. The compound is supplied as a lyophilized solid for research use and has no identified natural source in the body.
Activity is mediated mainly through the growth hormone secretagogue receptor, now generally called the ghrelin receptor or GHS-R1a. Binding at this G-protein-coupled receptor triggers phospholipase C signaling, calcium mobilization, and release of growth hormone from pituitary somatotrophs. Reports describe less pronounced stimulation of adrenocorticotropic hormone and prolactin compared with earlier secretagogues such as hexarelin or GHRP-6. Selectivity figures vary between assay systems, so the degree of separation from other secretagogues is an area of ongoing comparison rather than a fixed constant.
Research peptides such as ipamorelin are commonly supplied as lyophilized powder and characterized by analytical certificates. Reversed-phase high-performance liquid chromatography is used to estimate purity by ultraviolet absorbance, while mass spectrometry confirms molecular identity and detects sequence-related impurities. Counterion content, water content, and residual synthesis reagents can affect the reported mass balance. A certificate of analysis may list a purity percentage, but that number depends on the analytical method and the definition of impurity peaks. Independent verification is often recommended because research supply chains vary in quality control practices.
Storage recommendations for ipamorelin usually focus on temperature, moisture, and light. Lyophilized powder is typically held at or below minus twenty degrees Celsius in a desiccated container protected from light. Reconstituted solutions are often aliquoted and stored at minus eighty degrees Celsius to reduce repeated freeze-thaw cycles, which can promote aggregation or degradation. The optimal buffer and pH depend on the specific assay, and no single condition applies to every experimental context. Peptide stability should be assessed with time-point measurements rather than assumed from general handling rules.
In the scientific literature, ipamorelin appears mainly in preclinical studies, receptor binding assays, and reviews of growth hormone secretagogues. Authors often discuss its selectivity profile alongside limitations such as small sample sizes, short study durations, and differences between species. Some papers examine pharmacokinetics and clearance, but human data are limited and not sufficient to define general clinical effects. Regulatory discussion treats the compound as an investigational or research substance rather than an approved therapy in most jurisdictions. Open questions include oral bioavailability, long-term endocrine effects, and whether selectivity observed in animals persists in humans.
== Signal transduction pathway == A glucagon receptor, upon binding with the signaling molecule glucagon, initiates a signal transduction pathway that begins with the activation of adenylate cyclase, which in turn produces cyclic AMP (cAMP). Protein kinase A, whose activation is dependent on the increased levels of cAMP, is responsible for the ensuing cellular response in the form of protein kinase 1 and 2. The ligand-bound glucagon receptor can also initiate a concurrent signaling pathway that is independent of cAMP by activating phospholipase C. Phospholipase C produces DAG and IP3 from PIP2, a phospholipid phospholipase C cleaves off of the plasma membrane. Ca2+ stores inside the cell release Ca2+ when its calcium channels are bound by IP3.
=== 2005–present: A new look === On 17 December 2005, the organisation obtained a new corporate identity. The previous NOS logo was in use for 36 years and featured the company's initials in lowercase, with round and obtuse angles. The O of the logo is painted red and represents "the hub of 360-degree reportage". The new logo was designed by graphic designers Lambie-Nairn, complete with new idents for its television programmes. The typeface was Gotham. Its news-gathering operations also changed, with the news departments for radio, television, and internet merging and reorganising into different sections (NOS, NOS News, NOS Sports, with NOS Events added in 2009). An announcement made by the Minister of Education in 2007 confirmed a further division of the NOS. The umbrella group Publieke Omroep, managed by the NOS was to be legally separated and renamed Nederlandse Publieke Omroep (Netherlands Public Broadcasting). The move was to ensure the NOS operated under the same conditions as its fellow broadcasting associations, as well as transferring membership of the European Broadcasting Union. In 2009, the NOS became fully independent and now has its own Board of Directors. Since 2012, the NOS has got a new style and layout and, for some programs, other titles. The biggest change is visible in the 8 o'clock NOS Journaal (traditionally the most viewed and therefore the most important news broadcast of the NOS), where they adopted a standing presentation.
Group 1.4.1 is associated with connective tissue disorders including scleroderma (with scleroderma being the most common disorder with an estimated 8-19% of scleroderma patients having PAH), lupus, mixed connective tissue disease, rheumatoid arthritis, and Sjogren's syndrome. Group 1.4.2 is associated with HIV, group 1.4.3 associated with portal hypertension (commonly due to liver cirrhosis), group 1.4.4 associated with congenital heart disease, and 1.4.5 associated with schistosomiasis. In areas where schistosomiasis is prevalent, PAH due to schistosomiasis is believed to be the most prevalent variant of PAH. It is estimated that 5-8% of people with schistosomiasis with liver fibrosis have PAH. Group 1.5 PAH is the variant of PAH that is responsive to vasodilators (calcium channel blockers). The response to vasodilation by definition includes a decrease in the pulmonary arterial pressure of 10 mmHg or more to less than 40 mmHg without a decline in the cardiac output upon infusion of nitrous oxide(vasodilator) during the right heart catheterization. Vasodilator responsive PAH is estimated to be 6.8% of PAH patients in one study, and it is associated with a more favorable prognosis. Group 1.6 PAH includes PAH with features of venous or capillary involvement and is thought to comprise 5-10% of PAH cases. Two conditions, pulmonary veno-occlusive disease and pulmonary capillary hemangiomatosis comprise this category. The subcategory is characterized by severe hypoxemia, capillary congestion and prominent post-capillary venule thickening.

Obese individuals are predisposed to developing osteoarthritis, not only due to the excess mechanical load, but also due to the excess expression of soluble factors, that is, leptin and pro-inflammatory cytokines, which contribute to joint inflammation and cartilage destruction. As such, obese individuals are in an altered state, due to a metabolic insufficiency, which requires specific nutritional treatment capable of normalising the leptin production and reducing the systematic low-level inflammation, in order to reduce the harmful impact of these systematic mediators on the joint health.

Emerging technologies are novel technologies whose development or practical applications are still largely unrealized. They include nanotechnology, biotechnology, robotics, 3D printing, and blockchains. In 2005, futurist Ray Kurzweil claimed the next technological revolution would rest upon advances in genetics, nanotechnology, and robotics, with robotics being the most impactful of the three technologies. Genetic engineering will allow far greater control over human biological nature through a process called directed evolution. Some thinkers believe that this may shatter humanity's sense of self, and they have urged for renewed public debate exploring the issue more thoroughly; others fear that directed evolution could lead to eugenics or extreme social inequality. Nanotechnology will grant the ability to manipulate matter "at the molecular and atomic scale", which could allow humanity to reshape itself and its environment in fundamental ways. Nanobots could be used within the human body to destroy cancer cells or form new body parts, blurring the line between biology and technology. Autonomous robots have undergone rapid progress, and they are expected to replace humans at many dangerous tasks, including search and rescue, bomb disposal, firefighting, and war. Estimates on the advent of artificial general intelligence vary, but half of machine learning experts surveyed in 2018 believe that AI will "accomplish every task better and more cheaply" than humans by 2063, and automate all human jobs by 2140.
== Preparation and manufacturing == The simplest method for preparing ceramic superconductors is a solid-state thermochemical reaction involving mixing, calcination and sintering. The appropriate amounts of precursor powders, usually oxides and carbonates, are mixed thoroughly using a Ball mill. Solution chemistry processes such as coprecipitation, freeze-drying and sol–gel methods are alternative ways for preparing a homogeneous mixture. These powders are calcined in the temperature range from 1,070 to 1,220 K (800 to 950 °C) for several hours. The powders are cooled, reground and calcined again. This process is repeated several times to get homogeneous material. The powders are subsequently compacted to pellets and sintered. The sintering environment such as temperature, annealing time, atmosphere and cooling rate play a very important role in getting good high-Tc superconducting materials. The YBa2Cu3O7−x compound is prepared by calcination and sintering of a homogeneous mixture of Y2O3, BaCO3 and CuO in the appropriate atomic ratio. Calcination is done at 1,070 to 1,220 K (800 to 950 °C), whereas sintering is done at 1,220 K (950 °C) in an oxygen atmosphere. The oxygen stoichiometry in this material is very crucial for obtaining a superconducting YBa2Cu3O7−x compound. At the time of sintering, the semiconducting tetragonal YBa2Cu3O6 compound is formed, which, on slow cooling in oxygen atmosphere, turns into superconducting YBa2Cu3O7−x. The uptake and loss of oxygen are reversible in YBa2Cu3O7−x.
== Motivation == Researcher Frederick Banting famously refused to put his name on the patent after discovering insulin in 1923. The original patent for insulin was later sold by his collaborators for just $1 to the University of Toronto in an effort to make it as available as possible. Despite this, for various reasons, there remains no generic version of insulin available in the US. Insulin remains controlled by a small number of large pharmaceutical companies and sold at prices unaffordable to many who rely on it to live, particularly those without insurance. This lack of availability has led to fatalities, such as Alec Smith, who died in 2017 due to lack of insulin. The Open Insulin Project is motivated by the urgent need to protect the health of those with diabetes regardless of their economic or employment status by developing low-cost methods for insulin production available for anyone to use.
== International rules series == Fletcher served as the Australia international rules football team's goalkeeper for five series: the 2005 series, the 2006 series, the 2010 series, the 2014 series, and the 2015 series. Along with Barry Hall, Fletcher was one of Australia's co-captains for the 2006 series. He is regarded as Australia's greatest keeper throughout the series. He retired from top-level football with a final International Rules match, at the 2015 series in Ireland.

=== Synthesis === For the synthesis of alprazolam the same method can be used as for triazolam, except that it starts from 2-amino-5-chlorobenzophenone. However, an alternative easier synthesis starting with 2,6-dichloro-4-phenylquinoline has been suggested, in which it reacts with hydrazine giving 6-chloro-2-hydrazino-4-phenylquinoline. Boiling the mixture with triethyl orthoacetate results in cyclization with the formation of the triazole ring. The product undergoes oxidative degradation in the presence of periodate and ruthenium dioxide in acetone solution, giving 2-[4-(3'-methyl-1,2,4-triazolo)]-5-chlorobenzophenone. Oxy-methylation with formaldehyde results in a product that is treated with phosphorus tribromide when 2-[4-(3'-methyl-5'-bromomethyl-1,2,4-triazolo)]-5-chlorobenzophenone is obtained. By substituting the bromine atom with an amino group conferred by ammonia, it forms alprazolam triazolobenzophenone, following which an intramolecular heterocyclization takes place to obtain alprazolam.

It has not been approved as a therapeutic by major regulators, and the human trial record is small and dated. Material available today is mostly sold as a research chemical for laboratory use. Approval and restriction status varies by country.
It binds the ghrelin receptor, known as the growth hormone secretagogue receptor type 1a. Activation of that receptor triggers signaling that leads to growth hormone release. Selectivity for growth hormone over cortisol and prolactin is the property most often cited for this compound.
Published estimates place the circulating half-life at roughly two hours in humans. Clearance involves peptidase activity and renal handling. Because the peptide is short-lived, release patterns after administration are usually described as pulsatile rather than sustained.
It is a synthetic pentapeptide in the growth hormone secretagogue family. The chain contains five residues, two of which are non-natural amino acids.
